Insilico Reports Early ISM6331 Responses in Mesothelioma

Oct 27, 2026
Insilico Medicine reported a 40% objective response rate at the 130 mg dose of ISM6331 in a small group of participants with mesothelioma.

Insilico Medicine said in a press release that its experimental cancer drug ISM6331 produced a 40% objective response rate among five evaluable participants with mesothelioma who received 130 mg once daily. The preliminary Phase 1 results were presented at the ESMO Congress 2026 in Madrid.

Across doses from 30 mg to 130 mg, four of 22 evaluable participants with mesothelioma responded to treatment, an objective response rate of 18.2%. The disease control rate at the first tumor assessment was 68.2%, while three of the four partial responses were continuing at the August 14 data cutoff.

The trial enrolled 36 people with advanced solid tumors across doses from 15 mg to 130 mg. No dose limiting toxicities or treatment related discontinuations were reported, while Grade 3 treatment related adverse events occurred in 8.3% of participants. Insilico designed the oral TEAD1 through TEAD4 inhibitor using its Chemistry42 generative AI platform and plans to complete Phase 1 dose selection before a randomized Phase 2a study in mesothelioma.
